Trinsicon Intra Labs India is a medication for the treatment of erectile dysfunction. This drug restores impaired erectile function and provides a natural response to sexual stimulation. Sildenafil is a potent and selective inhibitor of cGMP-specific phosphodiesterase type 5 (PDE5) which is responsible for the breakdown of cGMP in the cavernous body. This medicine has no direct relaxing effect on corpus cavernosum, but enhances the relaxing effect of active nitric oxide in this tissue. When sexually aroused the local release of NO under the influence of sildenafil leads to the inhibition of PDE-5 and increase of cGMP in the cavernous body, resulting in relaxation of smooth muscles and increasing blood flow to the cavernous body. Sildenafil may cause mild and transient violation of distinguishing colors (blue / green). Alleged violations of the mechanism of color vision is PDE6 oppression, which is involved in the transmission of light in the retina.

How should I take Trinsicon?

Use this medicine as directed on the label, or as your doctor has prescribed. Do not use the medicine in larger amounts or for longer than recommended.

Never take more than the recommended dose of multivitamins with iron. Avoid taking any other multivitamin product within 2 hours before or after you take multivitamins with iron. Taking similar vitamin products together at the same time can result in a vitamin overdose or serious side effects.

Many multivitamin products also contain minerals such as calcium, magnesium, potassium, and zinc. Minerals (especially taken in large doses) can cause side effects such as tooth staining, increased urination, stomach bleeding, uneven heart rate, confusion, and muscle weakness or limp feeling. Read the label of any multivitamin product you take to make sure you are aware of what it contains.

You may take the medicine with food if it upsets your stomach.

The chewable tablet must be chewed or allowed to dissolve in the mouth before swallowing.

Measure liquid medicine with the dosing syringe provided, or with a special dose-measuring spoon or medicine cup. If you do not have a dose-measuring device, ask your pharmacist for one.

Liquid or powder multivitamin may sometimes be mixed with water, fruit juice, or infant formula (but not milk or other dairy products). Follow the mixing directions on the medicine label.

Do not crush, chew, break, or open an extended-release capsule or tablet. Swallow it whole.

It is important to take multivitamins with iron regularly to get the most benefit.

Store in the original container at room temperature away from moisture and heat. Do not allow the liquid to freeze.

After oral administration Trinsicon Intra Labs India is rapidly absorbed. The absolute bioavailability averages 40% (25-63%). After a single oral dose in 100 mg Cmax is 18 ng / ml and is reached in the fasting during 30-120 min. When taking sildenafil in combination with fatty food the intake rate decreases; Tmax increased by 60 min and Cmax decreased by an average of 29%. Vd of sildenafil at steady state an average is 105 liters. Sildenafil and its major circulating N-desmethyl metabolite approximately 96% bound to plasma proteins. Protein binding does not depend on the total concentration of sildenafil. Less than 0.0002% of the dose (mean 188 ng) were detected in semen up to 90 min after administration of sildenafil.

Trinsicon Intra Labs India is metabolized primarily by the action of microsomal liver isoenzymes CYP3A4 (major route) and CYP2C9.

The total clearance of sildenafil from the body is equal to 41 l / h, T1/2 in the terminal phase is 3-5 hours. After oral administration, sildenafil is displayed in the form of metabolites mainly in the faeces (approximately 80% of dose) and to a lesser extent in the urine (approximately 13% of dose).

In elderly patients (aged 65 and older) sildenafil clearance is reduced, and the concentration of free active substance in the plasma by approximately 40% higher than its concentration in young (18-45 years) patients.

In renal insufficiency, mild (creatinine clearance 50-80 ml / min) and medium (CC 30-49 ml / min) the severity of the pharmacokinetic parameters of sildenafil after oral administration of a single dose (50 mg) did not change. In renal failure, severe (CC < 30 mL / min), sildenafil clearance is reduced, resulting in approximately twofold increase in AUC (100%) and Cmax (88%) compared with those with normal indices of renal function in patients of the same age group.

In patients with liver cirrhosis (class A and B in Child-Pugh), sildenafil clearance is reduced, which leads to an increase in AUC (84%) and Cmax (47%) compared with those with normal parameters of liver function in patients of the same age group.
