Description: Dantrium Capsules has a direct action on skeletal muscle. It uncouples muscular contraction from excitation, probably by interfering with calcium release from sarcoplasmic reticulum.

Pharmacokinetics:

Absorption: Slowly and almost completely absorbed from the GI tract after oral admin.

Distribution: Extensively bound to plasma proteins.

Metabolism: Mainly hepatically metabolised to the hydroxylated metabolites.

Excretion: Mainly excreted in the urine as metabolites with a small amount of unchanged Dantrium Capsules; some is excreted in the bile. Elimination half-life: about 9 hr.

How should I take Dantrium Capsules?

Dantrium Capsules may be taken with or without food or on a full or empty stomach. However, if your doctor tells you to take the medicine a certain way, take it exactly as directed.

Take Dantrium Capsules only as directed by your doctor. Do not take more of it and do not take it more often than your doctor ordered.

Dosing

The dose of Dantrium Capsules will be different for different patients. Follow your doctor's orders or the directions on the label. The following information includes only the average doses of Dantrium Capsules. If your dose is different, do not change it unless your doctor tells you to do so.

The amount of medicine that you take depends on the strength of the medicine. Also, the number of doses you take each day, the time allowed between doses, and the length of time you take the medicine depend on the medical problem for which you are using the medicine.

Missed Dose

If you miss a dose of Dantrium Capsules, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular dosing schedule. Do not double doses.

Storage

Store the medicine in a closed container at room temperature, away from heat, moisture, and direct light. Keep from freezing.

Keep out of the reach of children.

Do not keep outdated medicine or medicine no longer needed.

Dantrium Capsules administration

Administration of drug is important to know because the drug absorption and action varies depending on the route and time of administration of the drug. A medicine is prescribed before meals or after meals or along with meals. The specific timing of the drug intake about food is to increase its absorption and thus its efficacy. Few work well when taken in empty stomach and few medications need to be taken 1 or 2 hrs after the meal. A drug can be in the form of a tablet, a capsule which is the oral route of administration and the same can be in IV form which is used in specific cases. Other forms of drug administration can be a suppository in anal route or an inhalation route.

Dantrium Capsules pharmacology

Pharmacokinetics of a drug can be defined as what body does to the drug after it is taken. The therapeutic result of the medicine depends upon the Pharmacokinetics of the drug. It deals with the time taken for the drug to be absorbed, metabolized, the process and chemical reactions involved in metabolism and about the excretion of the drug. In isolated nerve-muscle preparation, Dantrium Capsules has been shown to produce relaxation by affecting the contractile response of the skeletal muscle at a site beyond the myoneural junction, directly on the muscle itself. In skeletal muscle, Dantrium Capsules dissociates the excitation-contraction coupling, probably by interfering with the release of Ca++ from the sarcoplasmic reticulum. This effect appears to be more pronounced in fast muscle fibers as compared to slow ones, but generally affects both. A central nervous system effect occurs, with drowsiness, dizziness, and generalized weakness occasionally present. Although Dantrium Capsules does not appear to directly affect the CNS, the extent of its indirect effect is unknown. The absorption of Dantrium Capsules after oral administration in humans is incomplete and slow but consistent, and dose-related blood levels are obtained. The duration and intensity of skeletal muscle relaxation is related to the dosage and blood levels. The mean biologic half-life of Dantrium Capsules in adults is 8.7 hours after a 100-mg dose. Specific metabolic pathways in the degradation and elimination of Dantrium Capsules in human subjects have been established. Metabolic patterns are similar in adults and pediatric patients. In addition to the parent compound, Dantrium Capsules, which is found in measurable amounts in blood and urine, the major metabolites noted in body fluids are the 5-hydroxy analog and the acetamido analog. Since Dantrium Capsules is probably metabolized by hepatic microsomal enzymes, enhancement of its metabolism by other drugs is possible. However, neither phenobarbital nor diazepam appears to affect Dantrium Capsules metabolism.

Clinical experience in the management of fulminant human malignant hyperthermia, as well as experiments conducted in malignant hyperthermia susceptible swine, have revealed that the administration of intravenous Dantrium Capsules, combined with indicated supportive measures, is effective in reversing the hypermetabolic process of malignant hyperthermia. Known differences between human and swine malignant hyperthermia are minor. The prophylactic administration of oral or intravenous Dantrium Capsules to malignant hyperthermia susceptible swine will attenuate or prevent the development of signs of malignant hyperthermia in a manner dependent upon the dosage of Dantrium Capsules administered and the intensity of the malignant hyperthermia triggering stimulus. Limited clinical experience with the administration of oral Dantrium Capsules to patients judged malignant hyperthermia susceptible, when combined with clinical experience in the use of intravenous Dantrium Capsules for the treatment of malignant hyperthermia and data derived from the above cited animal model experiments, suggests that oral Dantrium Capsules will also attenuate or prevent the development of signs of human malignant hyperthermia, provided that currently accepted practices in the management of such patients are adhered to; intravenous Dantrium Capsules should also be available for use should the signs of malignant hyperthermia appear.
