The exact mechanism of action is not known. Dexedrine Spansule Sustained-Release Capsules stimulates the release of norepinephrine from central adrenergic receptors. At higher dosages, it causes release of dopamine from the mesocorticolimbic system and the nigrostriatal dopamine systems by reversal of the monoamine transporters. Dexedrine Spansule Sustained-Release Capsules may also act as a direct agonist on central 5-HT receptors and may inhibit monoamine oxidase (MAO). In the periphery, amphetamines are believed to cause the release of noradrenaline by acting on the adrenergic nerve terminals and alpha- and beta-receptors. Modulation of serotonergic pathways may contribute to the calming affect.

Do not take Dexedrine Spansule Sustained-Release Capsules late in the day because it may cause sleep problems (insomnia).
Do not crush, chew, break, or open an extended-release capsule. Swallow it whole.
Measure liquid medicine with a special dose-measuring spoon or medicine cup. If you do not have a dose-measuring device, ask your pharmacist for one.
Your doctor will need to check your progress while you are using Dexedrine Spansule Sustained-Release Capsules.
This medicine can cause unusual results with certain medical tests. Tell any doctor who treats you that you are using Dexedrine Spansule Sustained-Release Capsules.
Store at room temperature away from moisture, heat, and light.

Amphetamines are noncatecholamine, sympathomimetic amines with CNS stimulant activity. Peripheral actions include elevations of systolic and diastolic blood pressures and weak bronchodilator and respiratory stimulant action.
There is neither specific evidence that clearly establishes the mechanism whereby amphetamines produce mental and behavioral effects in children, nor conclusive evidence regarding how these effects relate to the condition of the central nervous system.
Dexedrine Spansule Sustained-Release Capsules Capsules are formulated to release the active drug substance in vivo in a more gradual fashion than the standard formulation, as demonstrated by blood levels. The formulation has not been shown superior in effectiveness over the same dosage of the standard, noncontrolledrelease formulations given in divided doses.
The pharmacokinetics of the tablet and sustained-release capsule were compared in 12 healthy subjects. The extent of bioavailability of the sustained-release capsule was similar compared to the immediaterelease tablet. Following administration of three 5-mg tablets, average maximal Dexedrine Spansule Sustained-Release Capsules plasma concentrations (Cmax) of 36.6 ng/mL were achieved at approximately 3 hours. Following administration of one 15-mg sustained-release capsule, maximal Dexedrine Spansule Sustained-Release Capsules plasma concentrations were obtained approximately 8 hours after dosing. The average Cmax was 23.5 ng/mL. The average plasma T½ was similar for both the tablet and sustained-release capsule and was approximately 12 hours. In 12 healthy subjects, the rate and extent of Dexedrine Spansule Sustained-Release Capsules absorption were similar following administration of the sustained-release capsule formulation in the fed (58 to 75 gm fat) and fasted state.
