Actions of Loxitane IM in details
Description: Loxitane IM, a dibenzoxazepine antipsychotic, antagonises central dopaminergic action by blocking postsynaptic mesolimbic D1 and D2 receptors in the brain. It also has serotonin 5-HT2 inhibiting activity.
Pharmacokinetics:
Absorption: Absorbed readily from the GI tract; peak plasma concentrations within 1-2 hr.
Distribution: Widely distributed; crosses the placenta and distributes into breast milk.
Metabolism: Very rapidly and extensively hepatic. Major metabolites: 7- and 8-hydroxyloxapine.
Excretion: Mainly via urine (as conjugated metabolites); via faeces (small amounts as unconjugated metabolites). 12-19 hr (terminal elimination half-life).
How should I take Loxitane IM?
Take Loxitane IM only as directed by your doctor. Do not take more of it, do not take it more often, and do not take it for a longer time than your doctor ordered. To do so may increase the chance of side effects.
Dosing
The dose of Loxitane IM will be different for different patients. Follow your doctor's orders or the directions on the label. The following information includes only the average doses of Loxitane IM. If your dose is different, do not change it unless your doctor tells you to do so.
The amount of medicine that you take depends on the strength of the medicine. Also, the number of doses you take each day, the time allowed between doses, and the length of time you take the medicine depend on the medical problem for which you are using the medicine.

- Adults—At first, 20 to 50 milligrams (mg) per day, divided and given in two to four doses per day. Your doctor may increase your dose if needed. However, the dose is usually not more than 250 mg per day.
- Children—Use and dose must be determined by your doctor.

Missed Dose
If you miss a dose of Loxitane IM, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular dosing schedule. Do not double doses.
Storage
Store the medicine in a closed container at room temperature, away from heat, moisture, and direct light. Keep from freezing.
Keep out of the reach of children.
Do not keep outdated medicine or medicine no longer needed.
Ask your healthcare professional how you should dispose of any medicine you do not use.

Follow all directions on your prescription label. Your doctor may occasionally change your dose to make sure you get the best results. Do not take this medicine in larger or smaller amounts or for longer than recommended.
Take Loxitane IM with a full glass of water.
You may not start feeling better right away when you start taking Loxitane IM. For best results, keep using the medication as directed. Talk with your doctor if your symptoms do not improve during treatment.

Pharmacodynamics
Pharmacologically, Loxitane IM (Loxitane IM (Loxitane IM (Loxitane IM succinate) succinate) succinate) is an antipsychotic for which the exact mode of action has not been established. However, changes in the level of excitability of subcortical inhibitory areas have been observed in several animal species in association with such manifestations of tranquilization as calming effects and suppression of aggressive behavior.
In normal human volunteers, signs of sedation were seen within 20 to 30 minutes after administration, were most pronounced within one and one-half to three hours, and lasted through 12 hours. Similar timing of primary pharmacologic effects was seen in animals.
Absorption, Distribution, Metabolism, and Excretion
Absorption of Loxitane IM (Loxitane IM (Loxitane IM (Loxitane IM succinate) succinate) succinate) following oral or parenteral administration is virtually complete. The drug is removed rapidly from the plasma and distributed in tissues. Animal studies suggest an initial preferential distribution in lungs, brain, spleen, heart, and kidney. Loxitane IM (Loxitane IM (Loxitane IM (Loxitane IM succinate) succinate) succinate) is metabolized extensively and is excreted mainly in the first 24 hours. Metabolites are excreted in the urine in the form of conjugates and in the feces unconjugated.
